Abstract Objective 'To investigate the proteetive elfeets of leonurine( LE0 )on pyroptosis induced by oxidized low.density lipoprotein( ox-LDL) in THP-1-derived maerophages. Vethods M'T'T assay was used to detect the viability of'THP-1 macrophages/'THP-1-derived foam cells after treatment with diflerent coneentrations of LEO. 0il Red Ostaining was used to assess the intracellular lipid accumulation, meanwhile the contents of intracellular totalcholesterl('TC ), triglycerides ( TG ), interleukin-1B ( 1-1B), interleukin-18 ( IL-18), and lactate dehydrogenase ( LDH) in the cell supernatan were detected. Westerm blot was used to detect the expression level of proteins relatedto cellular pyrolysis. The level of reactive oxygen species ( ROS) was determined by flow cytometry. Results As theconeentration of LE0 inereased, there was a positive correlation between cell activity and LE0 concentration. LEOintervention decreased the intracellular 'TC and 'TG levels, and reduced the intracellular lipid accumulation. LEOdecreased the expression of cellular pyroptosis-related proteins, including NLRP3 ,N-GSDMD,Cleaved Caspase-1. Atthe same time, treatment with LE0 reversed the ox-LDL-induced inerease in ROS production. Conclusion LEOdecreases lipid accumulation in 'THP-1 macrophages induced by ox-LDL and inhibits the pyroptosis induced by ox-LDL in THP-1-derired macrophage.
