Abstract Objective To study the effect of excretory products of Clonorchis sinensis ( Cs.ESP) and crude antigen of crude-antigen of Clonorchis sinensis ( Cs. CA ) on the activation of primary cultured hepatic stellate cells ( HSCs) from mice. Methods The primary HSCs from mice were separated, identified and cultured in vitro. HSCs were divided into the Cs. ESP stimulation group, Cs. CA stimulation group, and a blank control group. The mRNA levels of Collagen I and α-smooth muscle actin ( α-SMA) , the activation indicators of HSCs were detected by RT-PCR, and the protein expressions of Collagen I and α-SMA were detected by Western blot. Results The mRNA and protein expression levels of CollagenⅠ and α-SMA in the Cs.ESP stimulation group were up-regulated compared with those in the Cs. CA stimulation group and blank control group, showing a statistically significant difference ( P < 0. 05) . There was no significant difference in the mRNA and protein expression levels of CollagenⅠ and α-SMA between Cs.CA stimulation group and blank control group ( P>0.05) . Conclusion Cs. ESP promotes the activation of HSCs in vitro, while Cs.CA has no obvious effect on the activation of HSCs. This study provides a reference for further research on the cellular and molecular mechanisms of liver fibrosis induced by Cs.ESP.
